MS-347a is a fungal secondary metabolite isolated from Aspergillus sp. that irreversibly inhibits smooth muscle myosin light chain kinase (MLCK) with micromolar potency. The compound suppresses phosphorylation of myosin light chain by binding to the catalytic domain of MLCK and inhibits both calmodulin-dependent and calmodulin-independent enzyme activity. MS-347a attenuates sustained Ca2+-dependent exocytosis and inositol trisphosphate signaling without broadly inhibiting other cyclic nucleotide-dependent kinases. It has been used as a mechanistic probe to dissect MLCK-mediated actomyosin regulation and calcium-controlled secretion pathways.
