NG-012 is a secondary metabolite isolated from Penicillium verruculosum that enhances nerve growth factor (NGF)-induced neurite outgrowth in PC12 cells. Similar to NG-011, it does not mimic NGF directly but potentiates NGF-dependent signaling pathways, thereby increasing neuronal differentiation responses. Structural studies revealed that NG-012 belongs to a macrocyclic polylactone class of fungal metabolites. It serves as a neurobiology research tool to investigate NGF-TrkA signaling, neuronal differentiation mechanisms, and neurotrophic pathway modulation.
