Details
PF1052 is a tetramic acid肪earing cis-decalin natural product originally isolated from Phoma sp. and structurally related to vermisporin. Although not active in lipid droplet accumulation assays, PF1052 was later identified as a potent inhibitor of neutrophil recruitment in transgenic zebrafish and murine chemotaxis models. It suppresses pseudopod formation and neutrophil motility without directly inhibiting PI3K activation or AKT phosphorylation, suggesting modulation of downstream cytoskeletal signaling. Total synthesis studies confirmed its absolute configuration and established efficient access to this scaffold. PF1052 therefore serves as a valuable chemical probe for studying inflammatory cell migration and cytoskeletal dynamics.
